So whenever we do a genetic test there can be three answers that we get: it could be clearly negative so sometimes variants are found that are benign or likely benign ― they're non-disease-causing variants. Usually these are so common in the population. They're not even going to be reported out on your test report. A result could be clearly positive, a pathogenic or likely pathogenic or likely disease-causing variant and this is typically a variant where the gene change in the gene truly causes the gene not to work. Maybe it's been reported in the literature and someone who has Loeys-Dietz syndrome. In this instance we can use medical guidelines to help take care of you. We can clearly test family members and say who has disease or who does not have disease.

And then sometimes when we do genetic testing we can get an answer called a variant of uncertain significance, so this is a gene change where we're not 100 sure it truly causes disease, we're not 100 sure we can blow it off that it's a non-disease-causing variant and a lot of Loeys-Dietz variants kind of live in this center section. We're pretty good at interpreting VUSs and say it's likely in the pathogenic versus non-pathogenic category and sometimes we do that by tracking it in family members, by asking specialty groups to see a lot of patients whether they've seen this variant and only in a small, small percentage is it truly an unknown answer.

And over time as we test more and more family members, as we test more and more people there are possibilities that variants of uncertain significance get upgraded to pathogenic or could even get downgraded, so it's kind of a, for some of these VUS it's a work in progress.
