For, families that have like multiple generations of Loeys-Dietz syndrome, someone is asking a question that in the third generation it appears to be a bit more worse than the previous generations? Does it get worse over time?

So that's not known to be broadly true or predictable for Loeys-Dietz syndrome. There are some genetic diseases where that is known to be true with each successive generation. The condition is at high risk to get worse but that is a very unique circumstance based upon the mechanism that's causing the disease.

世代を重ねるごとに病態が悪化するかどうかは、LDSでははっきりとわかっていません。つまり、次世代の病態についての予測はできないということです。いくつかの遺伝性疾患では、病態悪化のリスクが高まることがわかっていますが、その要因は発症メカニズムにあるため、非常に特殊な状況でのみ起こる事象といえます。
That mechanism is not in play in Loeys-Dietz syndrome and therefore I don't think that that is caused by being in the next generation. I think it's just the type of variation that you can see at random. For example, it's possible that that more severely affected child could have a child with Loeys-Dietz syndrome who again shows mild features of the disease, so it's not getting worse in each successive generation.
